Evaluation of cytokine mRNA expression in vaccinated guinea pigs with foot-and-mouth disease type O inactivated vaccine

Abstract

Foot-and-mouth disease (FMD) is a severely contagious viral disease that mainly affects cloven-hoofed
livestock and wildlife. This study quantifies the cytokines mRNA expression of vaccinated guinea pigs with
FMD type O inactivated vaccine. Blood samples were collected from eight guinea pigs at 7 and 28 days after the
first vaccination. Extracted mRNAs were reverse-transcribed into cDNA and analyzed for quantification of IFN-
γ, TNF-α and IL-10 expression using relative real-time PCR assay. Our results showed that all of the genes were
upregulated. The expression of TNF-α and IL-10 genes significantly increased (P<0.05) in day 28th in
comparison to the day 7th post the first vaccination. It can be concluded that the vaccine induced immune
responses by increasing expression of the cytokines. Therefore, effects of DNA vaccines on immune system also
may be evaluated using these genes.

Abstract [French]

La fièvre aphteuse est une maladie virale fortement contagieuse qui touche principalement le bétail ongulé et la faune sauvage. Cette étude a pour but de quantifier l’expression ARNm des cytokines chez des cochons d’Inde vaccinés avec le type O inactivé du vaccin de la fièvre aphteuse. Les échantillons sanguins ont été prélevés aux 7e et 28e jours qui suivaient la première vaccination de 8 cochons d’Inde. Les ARNm isolés ont été soumis à une transcription inversée pour former des ADNc et analysés par PCR en temps réel relative afin de quantifier l’expression des cytokines IFN-γ, TNF-α et IL-10. Nos résultats montrent que l’expression de tous les gènes analysés est régulée à la hausse. L’expression des gènes codant le TNF-α et l’IL-10 augmente de façon significative (P<0.05) au 28e jour comparé au 7e jour après la première vaccination. On peut donc en conclure que la réponse immune induite par le vaccin est en partie due à une expression plus importante des cytokines étudiées. De ce fait, l’expression de ces gènes peut être utilisée comme marqueur spécifique de l’efficacité des vaccins d’ADN.
